The effect of r-telencephalin (NT) on release of [3H]acetylcholine ([3H]ACh) from primary cell cultures of embryonic chick brainstem was investigated. NT (1-100 nM) stimulated [3H]ACh release in a dose-dependent manner (ED50 = 6.4 nM). NT-stimulated [3H]ACh release was maximal (178.1 +/- 9.5% of basal release) at 30 min, declined to 112.4 +/- 7.2% at 90 min, and was no longer significant at 150 min. NT-stimulated release was blocked by (1) naloxone (10 microM), (2) methysergide (10 microM), (3) bacitracin (100 micrograms/ml), and (4) picrotoxin (10 microM). NT-stimulated [3H]ACh release was associated with a 2.5-fold increase in [3H]gamma-aminobutyric acid ([3H]GABA) release. To determine if NT-stimulated [3H]ACh release was associated with non-cholinergic (muscarinic) receptors, we tested NT for inhibition of [3H]ACh release stimulated by the muscarinic agonist oxotremorine (10 microM). Oxotremorine (10 microM) stimulated [3H]ACh release was blocked by NT (100 nM) in a dose-dependent manner (ED50 = 7.6 nM). The response to oxotremorine was blocked by methysergide (10 microM), bacitracin (100 micrograms/ml) and picrotoxin (10 microM). NT and oxotremorine were equipotent in inhibiting [3H]ACh release (ED50 = 16.9 nM).
